Výskyt a diverzita amphistomních motolic u skotu

The thesis deals with gastric woodlice of the family Paramphistomidae, which are common parasites of ruminants. Their immature stages can cause severe pathological changes in the host's digestive tract, and massive infection can lead to host death. Adult parasites cause infections of the rumen mucosa, resulting in lower production. The prevalence and diversity of these flukes is currently not systematically monitored. The aim of this study is to describe the occurrence and diversity of gastric flukes in cattle in the Czech Republic and Austria. The practical part consists of examination of dairy cattle faeces from different herds for the presence of eggs using standardised parasitological methods, optimization of a PCR method for direct detection of flukes from the faeces, genotyping of the flukes found, processing of the results using appropriate statistical methods, and comparison with published data.

The aim of the study is to describe the life cycle of a trematode species Anomalotrema koiae Gibson & Bray, 1984 (Digenea: Opecoelidae) which occures in three species of whelks (Buccinum undatum, B. glaciale and Plicifusus kroyeri) and Arctic staghorn sculpin (Gymnocanthus tricuspis) in littoral waters, mainly fjords, of the Svalbard archipelago to better understand the ecology of arctic marine habitats. In the study there is the first morphological description of larval stages of A. koiae, mainly the sporocysts and cercariae. For the purpose of the study, there were used both the molecular and morphological methods to analyse the data from the samples which were collected on Svalbard.
Determination of gastropods of the genus Radix, the intermediate hosts of trematodes

Lymnaeid snails of the genus Radix are common freshwater molluscs of Europe. Formerly, the following species have been reported from the Czech Republic according to the morphology of the shell: R. auricularia, R. peregra, R. ovata and R. ampla. However, recent studies involving molecular data revealed that species determination based on traditional approaches employing morphology of the shell and gonads can be more complicated, confusing and not fully reliable. The shape of the shell is affected by particular life conditions of the snails and proves to be highly variable. The characteristics that can be found on gonads are dependent on the reproductive phase of the snail, and sometimes also on the presence of trematode larval stages, which can cause parasitic castration. Recently, molecular taxonomy is the most reliable approach, especially when a combination of sequences of mitochondrial and nuclear DNA is used. As molecular analyses are relatively time-consuming and expensive, some authors try to find some additional morphological features that could be used for determination of Radix snails in the field, without the need of demanding equipment. The occurrence of Fascioloides magna in deers in South Bohemia

Giant liver fluke Fascioloides magna is the biggest represenative of flukes in the Czech Republic. This originally North American parasite was brought to Europe with wapiti deers in 1960s. Since that time Fascioloides magna has extended the area of its appearance and its host spectrum a lot. The first appearance of this parasite in the Czech Republic was noted for the first time at a fallow deer which was hunted down in Middle Bohemia in 1930. However in the early 1960s the number of positive findings started to increase at hoofed. In the last decade was observed the expansion of areas of appearance of this fluke in South Bohemia. That is why I decided to make examinations to evaluate the recent appearance of F. magna at deer family in the South Bohemia from March 2009 to January 2010. 74 samples of dropping of a deer from three different areas (Českobudějovicko, Třeboňsko and Šumava national park) were collected and examined and only in the sample from Šumava national park were proved the ovulums of F. magna. Then I examined 11 livers of deers which were hunted down in Šumava national park. In all these livers were found macroscopic changes. In all these examined livers was also finding of fluke F. magna in number from 1 to 72. From the reached results follows that the problems of appearance of F. magna is in the Czech Republic still actual and it is necessery to count with the extending area of appearance of this parasite in the future. Edification among hunting community and the use of information from trained people who make the primary examination of animals after hunting down are possible ways to catch up appearance and expansion of this serious parasite at hoofed.
